Rabbit Ear Ischemia

Rabbit ear ischemia is an experimental model in which blood flow to the ear or a defined tissue region is reduced or interrupted to investigate the effects of oxygen and nutrient deprivation. Vascular occlusion limits perfusion, alters cellular metabolism, and can produce tissue injury; restoring circulation may add oxidative stress and inflammation through ischemia-reperfusion mechanisms. In medicine, this model supports research on microvascular function, tissue survival, wound healing, angiogenesis, and reconstructive procedures. It also provides a controlled system for evaluating treatments intended to preserve perfusion, reduce tissue damage, or improve recovery after vascular compromise.

This protocol describes the induction of an ischemia-reperfusion (IR) model on mouse ear skin using magnet clamping. Using a custom-built intravital imaging model, we study in vivo inflammatory responses post-reperfusion. The rationale behind the development of this technique is to extend the understanding of how leukocytes respond to skin IR injury.

The present study demonstrates a highly reproducible animal model of acute regional myocardial ischemia and reperfusion injury in rabbits using a left mini-thoracotomy for survival cases or a midline sternotomy for non-survival cases.
