Unilateral Contusion Model

The unilateral contusion model is an experimental method that produces a controlled bruise or blunt-force injury on one side of a tissue or organ to study localized damage and recovery. A calibrated impact disrupts cells and small blood vessels, triggering inflammation, edema, and tissue remodeling at the injured site, while the uninjured side can provide an internal comparison. In medicine, this model helps researchers characterize injury progression, identify biological markers, and evaluate treatments aimed at reducing damage or improving repair. Its controlled, side-specific design supports clearer comparisons between injured and unaffected tissue in preclinical research.

2012

A reliable and repeatable way to produce a cervical unilateral spinal cord injury using the Infinite Horizon impactor is described. The method takes advantage of a custom designed frame and clamp to stabilize the spine. The standardized procedure and biomechanical injury parameters result in sufficient and sustained injuries.

A Contusion Model of Severe Spinal Cord Injury in Rats

A contusion model of severe spinal cord injury is described. Detailed pre-operative, operative and post-operative steps are described to obtain a consistent model.

The murine model of irreversible unilateral ureteric obstruction (UUO) is presented together with the model of reversible UUO in which the ureteric obstruction is relieved by anastomosis of the severed ureter into the bladder. These models enable the study of renal inflammation and scarring as well as tissue remodeling.

Source: Elzat, E. Y. Y., et al., Establishing a Mouse Contusion Spinal Cord Injury Model Based on a Minimally Invasive Technique. J. Vis. Exp. (2022).This video demonstrates the method for inducing a contusion injury at the T9 level spinal cord in a mouse model, using a spinal cord injury coaxial platform. A weight is dropped on the T9 level spinal cord that creates a contusion that immediately causes tissue discoloration and swelling.

A protocol for performing unilateral 6-OHDA lesions of the medial forebrain bundle in mice is described. This method has a low mortality rate (13.3 %) with 89% of the surviving animals showing >95% loss of striatal dopamine and 90.63±-4.02 % ipsiversive rotational bias towards the side of the lesion.
