Subclinical Atherogenesis

Subclinical atherogenesis is the early development of atherosclerotic arterial lesions before they produce symptoms or overt cardiovascular disease, making it an important target for prevention. It begins when endothelial dysfunction permits apoB-containing lipoproteins, particularly LDL, to enter and become retained in the arterial wall, where inflammatory signaling recruits immune cells that form foam cells and promote plaque growth and remodeling. In medicine, studying this silent phase helps researchers identify cardiovascular risk through biomarkers and vascular imaging, evaluate how hypertension, diabetes, smoking, and dyslipidemia accelerate disease, and assess interventions that may delay plaque progression and reduce later myocardial infarction or stroke.

We describe procedures to quantify and characterize atherosclerotic lesions in mouse models using precision sectioning of the aortic sinus and ascending aorta combined with histochemical and immunohistochemical analysis.

Transthoracic Speckle Tracking Echocardiography for the Quantitative Assessment of Left Ventricular Myocardial Deformation

Speckle tracking echocardiography is an emerging diagnostic imaging technique for the quantitative assessment of global and regional myocardial performance. Standard view echocardiographic motion images are recorded and deformation parameters are subsequently measured by automated continuous frame-by-frame tracking and motion analysis of speckles within the B-mode images of the myocardium.

Laser-assisted Microdissection (LAM) as a Tool for Transcriptional Profiling of Individual Cell Types

Here we present a protocol for laser-assisted microdissection of specific plant cell types for transcriptional profiling. While the protocol is suitable for different species and cell types, the focus is on highly inaccessible cells of the female germline important for sexual and apomictic reproduction in the crucifer genus Boechera.
