Manual Tracing

Manual tracing is a neuroscience technique for reconstructing the shape and organization of neurons or other neural structures from microscopy images, making cellular anatomy available for quantitative analysis. Researchers inspect image stacks or serial sections and use software or drawing tools to follow cell bodies, axons, dendrites, and branching points, recording their paths as digital annotations. The resulting reconstructions support measurements of neuronal morphology, connectivity, and spatial relationships, helping investigators compare brain regions, developmental stages, or disease-associated changes. Although labor-intensive, manual tracing remains valuable when image quality, labeling patterns, or complex anatomy challenge automated segmentation methods.

As manual dexterity is a prerogative mainly of primates, behavioral tasks have been developed in macaque monkeys. We present a method to collect cerebrospinal fluid (CSF) and to create a system which lacks CSF within the embryonic zebrafish brain ventricular system. This allows for further examination of CSF composition and its requirement during embryonic brain development.
