Microglial Depletion

Microglial depletion is the experimental removal or substantial reduction of microglia, the resident immune cells of the central nervous system, to investigate their roles in brain function and disease. Researchers commonly achieve it by inhibiting colony-stimulating factor 1 receptor (CSF1R), a survival-signaling pathway required by microglia, or through targeted genetic approaches; removing the treatment can allow repopulation from surviving precursor cells. In neuroscience, this method helps distinguish microglial contributions to neuroinflammation, synaptic remodeling, neurodegeneration, and neural injury. Comparing depleted and repopulated brains can reveal microglial functions and identify potential therapeutic strategies while highlighting effects caused by depletion itself.

Macrophages play a central role in homeostasis and pathology in many tissues. The protocol presented here describes methods for depleting macrophages in vivo, deriving polarized macrophages from bone marrow aspirates, and adoptively transferring macrophages into mice. These techniques allow determination of the role that polarized macrophages play in health and disease.

Microglia can influence neurons and other glia in culture by various non-cell autonomous mechanisms. Here, we present a protocol to selectively deplete microglia from primary neuronal cultures. This method has the potential to elucidate the role of microglial-neuronal interactions, with implications for neurodegenerative conditions where neuroinflammation is a hallmark feature.
