Nerve Fascicle Separation

Nerve fascicle separation is a microsurgical and anatomical technique for isolating the individual bundles of axons within a peripheral nerve, making their organization available for study or targeted manipulation. Under magnification, the operator exposes the nerve, follows connective tissue planes between fascicles, and gently separates them while preserving the perineurium and minimizing traction-related damage. In neuroscience, this approach supports studies of peripheral nerve anatomy, selective nerve repair and grafting, and the relationship between fascicular structure and functional pathways. Carefully separated fascicles can also provide tissue preparations for histological, electrophysiological, and neuroprosthetic research.

This video demonstrates a single-fiber recording of C-fiber nerve conduction in rats. An anesthetized rat is incised to expose the sciatic nerve, which provides sensory and motor supply to the lower limbs. The individual C-fibers are then exposed. Electrical stimulation is provided using stimulus electrodes implanted in the foot, and a response is recorded from a single nerve fiber.
