Stellate Cells

Stellate cells are star-shaped neurons with dendrites radiating from the cell body, a structure that supports the integration of signals across local neural circuits. Their branching dendrites receive input from multiple sources, while their axons transmit either excitatory or inhibitory signals depending on the cell type and brain region. In the cerebral cortex, stellate cells contribute to sensory processing, whereas cerebellar stellate cells help regulate activity within cerebellar microcircuits. Studying these neurons reveals how morphology, synaptic connectivity, and neurotransmitter release shape information processing, and it supports research into circuit function, development, and neurological disease.

Here we describe a method for the isolation of hepatic stellate cells from mouse liver. For stellate cell purification, mouse livers are digested in situ and in vitro by pronase-collagenase treatment prior to density gradient centrifugation. This technique yields highly pure hepatic stellate cells.

Here we show our method to isolate mouse epidermal neural crest stem cells (EPI-NCSC). Technique involves micro-dissecting whisker follicles, isolating the bulge and placeing it into tissue culture. EPI-NCSC start to emigrate from bulge explants onto the substratum within 3 - 4 days.
