Centrifugation

Centrifugation is a method of separating suspended particles from a liquid by spinning in a centrifuge at a high number of revolutions per minute.
&#160;The particles sediment at a rate proportional to the centrifugal force they experience.
After centrifugation, the particles settle at the bottom, while the supernatant liquid can be decanted.&#160;
When the components are of equal density, the heavier particles sediment first. However, among particles with equal mass, the denser particles sediment first.
In differential centrifugation, heavier particles are eliminated first at lower speeds. The centrifugation speed is increased stepwise until the target particles are isolated.
In density gradient centrifugation, the density of the solution increases toward the bottom of the tube.
Upon centrifugation, each component stops moving when it reaches a layer with equal density, resulting in separate bands.

Centrifugation is a separation technique based on differences in density or size. It is commonly used to separate solids from aqueous interferents. During centrifugation, the sample is placed in centrifugation tubes and spun at high angular velocity, which allows centrifugal force to act differentially on the different densities or masses of the components. After spinning, the supernatant liquid is decanted. Depending on the specific application, either the pellet or the supernatant is retained for further purification. Sometimes, the supernatant is subjected to additional rounds of centrifugation and downstream processing.
In principle, centrifugation separates particles based on differences in size and density. Larger particles tend to be heavier and tend to sediment first. If particles are similar in size, denser particles tend to sediment first due to higher sedimentation rates. In practice, complex mixtures such as cell lysates may not yield distinctly separated particles despite differences in density or size. To address this issue, researchers have devised a variety of centrifugation techniques.
The simplest centrifugation technique is differential centrifugation. Here, the particles to be separated have similar densities, so larger particles will sediment at lower speeds. The speed is increased stepwise until the target particles are isolated.
A more sophisticated method is equilibrium density gradient centrifugation. In this technique, the analyte is placed in a solution with a preformed density gradient or a solution that forms a density gradient during centrifugation. Here, the density of the solution increases towards the bottom of the tube, so the sedimentation rate of each analyte component decreases as it moves toward the bottom. When a component&#39;s density equals that of the solution, the centrifugal force acting on it becomes zero, and sedimentation ceases. As a result, each component is isolated in a layer equal to its density.

Centrifugation separates particles in a liquid by using differences in size and density. The sample is placed in centrifugation tubes and spun at high angular velocity. This creates centrifugal force, which makes the different components move at different rates.

After spinning, the supernatant, or liquid above the settled material, is decanted. In some cases, the pellet is kept for further purification. In other cases, the supernatant is kept and may be spun again for additional processing.

This method works because larger particles are usually heavier and sediment first. When particles are similar in size, denser particles tend to sediment first because they have higher sedimentation rates. Complex mixtures, such as cell lysates, may still not separate cleanly even when the components differ in size or density.

Researchers use different centrifugation methods to handle these mixtures. Differential centrifugation is the simplest approach. In this method, particles with similar densities are separated step by step by increasing the speed until the target particles are isolated.

A more advanced method is equilibrium density gradient centrifugation. In this technique, the analyte is placed in a solution with a preformed density gradient, or in one that forms a gradient during spinning. The solution becomes denser toward the bottom of the tube, so sedimentation slows as each component moves downward. When a component reaches a point where its density matches the solution, the centrifugal force on it becomes zero and it stops moving. Each component is then isolated in a layer that matches its density.

Understand and be prepared to use values specific to chemical processes: the mole, molar mass, molarity, and percent composition. EXPLANATION: During the early and mid-1800s, a tremendous amount of early research poured into building an understanding of the way that atoms combine. This work required analysis of the percent composition of different substances. Joseph Proust's work with tin oxides led him to publish his law of definite proportions noting that a given pure substance will have a constant ratio for the combination of elements making up the substance. Building on Proust's work, John Dalton published his law of multiple proportions and atomic theory noting that multiple patterns exist for the way that atoms combine, yet the combining atoms are invariable. Meanwhile Amedeo Avogadro hypothesizes that given equal pressures and temperatures, two equal volumes of gas will contain equal numbers of atoms. Acknowledging that the exact number of atoms was unknown in these gaseous volumes, the Latin word "mole" was used. "Mole" translates to "lump" recognizing the ambiguous quantity of gas. A number of attempts have been undertaken historically to quantify the mole. Robert Millikan's oil drop experiment revealed the change of a single electron, which could then be divided into Faraday's Constant for the charge of a mole of electrons. Dividing the charge of a mole of electrons by the charge of a single electron yields the number of electrons in a mole, $6.022\times10^{\wedge}23$. Students should be familiar with how to translate between the given quantities. Student's first use atomism/Dalton's atomic theory in third grade noting that matter is made up of particles too small to be seen. This the first time where students will undertake the process of counting these invisible particles. Electrolysis of water provides a tangible experience to demonstrate the constituent parts of water, though the definite proportions of water may not be easily seen due to the solubility of the gasses produced. The concept of relative mass can be explored by comparing small groups of like items. For example, relative masses of beans can be compared with pinto, white, and black beans. If bags are created with equal numbers of like beans, students can develop a concept of why the smallest bean will have a relative mass of 1, analogous to hydrogen.

Identify acids and bases as a special class of compounds with a specific set of properties. EXPLANATION: The concept of pH is first introduced in seventh grade as a chemical property of matter.
