Brain Nuclei

Brain nuclei are distinct clusters of neuronal cell bodies within the central nervous system that organize information and coordinate functions, including behavior. Each nucleus receives and integrates specific neural signals, then communicates with other brain regions through synaptic connections and neurotransmitter pathways to influence processes such as movement, motivation, learning, emotion, and sleep. In behavioral neuroscience, researchers examine nuclei using anatomical tracing, electrophysiology, imaging, and targeted manipulation to link circuit activity with observable actions. Studying these structures clarifies how distributed neural networks generate behavior and can reveal mechanisms underlying neurological and psychiatric disorders.

Neuronal Nuclei Isolation from Human Postmortem Brain Tissue

2008

The cellular heterogeneity of brain tissue poses a significant limitation for the study of epigenetic markings in chromatin because most assays lack single cell resolution. Neurons typically are intermingled with glia and other non-neuronal cells. We provide a protocol to extract and collect neuronal nuclei from human brain.

Isolation of Oxytocin Receptor-Rich Nuclei from a Neonatal Rat Brain

2025

This video demonstrates the isolation of oxytocin receptor-rich nuclei from the neonatal rat brain. Coronal slicing, followed by identification and precise punching of oxytocin receptor-rich brain nuclei, enables the effective extraction of oxytocin receptor-rich nuclei for further analysis.

High-Quality Brain and Bone Marrow Nuclei Preparation for Single Nuclei Multiome Assays

2023

The success of single-cell/single-nuclei transcriptomics and multi-omics largely depends on the quality of cells/nuclei. Therefore, isolating cells/nuclei from tissue and their purification must be highly standardized. This protocol describes the preparation of nuclei from the brain and bone marrow for downstream single-nuclei multiome assay.

Isolation of Nuclei from Fresh Frozen Glioma Tissues: A Method to Obtain Intact Nuclei from Glioma Tumor Samples

2023

This video describes the protocol to isolate single-nuclei from fresh frozen glioma tissue. The isolated single-nuclei can be used as specimens for sequencing techniques such as single-nuclei RNA (snRNA) sequencing and single-nuclei Assay for Transposase-Accessible Chromatin (snATAC) sequencing to obtain deeper insights into inter- and intra-tumor heterogeneity observed in glioma of the central nervous tissues.
