Csf Biomarker Status

CSF biomarker status describes whether specific biological markers are detected in cerebrospinal fluid and, when measured, whether their concentrations fall within or outside defined ranges. Researchers typically obtain CSF through lumbar puncture and analyze proteins, metabolites, inflammatory signals, or nucleic acids using biochemical or molecular assays; the resulting profile can reflect processes occurring in the central nervous system. In behavioral research, CSF biomarker status helps relate brain pathology, neuroinflammation, synaptic dysfunction, or neurodegeneration to changes in cognition, mood, motivation, and social behavior. It can support disease classification, participant stratification, prognosis, and evaluation of treatment-related biological effects.

We describe a high-throughput, multiplex, and targeted proteomic cerebrospinal fluid (CSF) assay developed with potential for clinical translation. The test can quantitate potential markers and risk factors for neurodegeneration, such as the apolipoprotein E variants (E2, E3 and E4), and measure their allelic expression.

We designed a cell-free receptor binding assay in order to estimate the binding of granulocyte-macrophage colony-stimulating factor (GM-CSF) to the receptors. It enables us to evaluate competitive inhibition of biotinylated GM-CSF binding to soluble GM-CSF receptor alpha by GM-CSF autoantibody with excellent reproducibility.

Here detailed protocols for culturing the murine myeloid precursor 32D/G-CSF-R cell line, performing viral infections, and carrying out proliferation and differentiation assays are presented.
