Large Volume Illuminator

A Large Volume Illuminator is an optical device that delivers controlled illumination across an extended three-dimensional region, making it useful when behavior must be observed throughout a large experimental space. Rather than concentrating light on a small focal plane, it directs excitation across the volume so moving organisms remain visible during imaging or behavioral recording. In behavioral research, this approach supports observation of freely moving subjects, improves alignment between illumination and video acquisition, and enables analysis of movement, posture, interactions, and responses to environmental or experimental stimuli. By expanding the observable field, large-volume illumination helps connect behavior with spatial context and experimental conditions.

High-resolution intravital imaging with enhanced contrast up to 120 µm depth in lymph nodes of adult mice is achieved by spatially modulating the excitation pattern of a multi-focal two-photon microscope. In 100 µm depth we measured resolutions of 487 nm (lateral) and 551 nm (axial), thus circumventing scattering and diffraction limits.
