Olfactory Discrimination

Olfactory discrimination is the ability to detect and distinguish among odors, allowing organisms to identify environmental cues, food, threats, and social signals. It depends on odor molecules activating different combinations of receptors in the olfactory epithelium, which generate distinct neural patterns that are processed through the olfactory bulb and higher brain regions; learning and prior experience can further shape behavioral responses. In behavioral research, olfactory discrimination tasks measure sensory perception, memory, attention, and decision-making by testing whether an animal or person can differentiate odor pairs or categories. These measures help clarify how sensory systems support adaptive behavior and how neurological or psychiatric conditions affect perception.

We present a simple and unbiased olfactory test in mice. With this protocol olfactory discrimination, preference, avoidance and sensitivity to a novel odor as compared to water can be assessed in single behavioral sessions. This method is indicated for a single experimenter and analysis is based on computer-assisted video processing.

This manuscript describes a protocol to examine the olfactory system of rodents. The olfactory habituation/dishabituation test will allow investigators to determine whether a mouse habituates to a repeatedly presented odor and whether the mouse demonstrates dishabituation when presented a novel odor.
