Tact Mand Echoic Sequelic

Tact, mand, echoic, and sequelic are verbal operants in behavior analysis, each defined by its controlling variables and consequences; together, they explain how language functions rather than simply what words mean. A mand occurs under a motivating operation and produces specific reinforcement, a tact is evoked by a nonverbal stimulus and typically receives generalized conditioned reinforcement, an echoic repeats a verbal model with point-to-point correspondence, and a sequelic follows a verbal stimulus without that correspondence. This framework supports functional communication training, language assessment, and individualized instruction by linking observable antecedents and consequences to verbal behavior.

RNA Isolation of Pseudomonas aeruginosa Colonizing the Murine Gastrointestinal Tract

A reliable method for the RNA isolation of Pseudomonas aeruginosa recovered from murine cecums is described. The RNA recovered is of sufficient quantity and quality for subsequent qPCR, transcription profiling, and RNA Seq experiments. This technique can be adapted for RNA isolation of other intestinal microbes.

Sepsis refers to a systemic inflammatory response syndrome resulting from a microbial infection, and can be simulated by a surgical technique termed cecal ligation and puncture (CLP). Here we describe a method to use CLP-induced animal model to screen medicinal herbs for therapeutic agents.

An approach to analyze the migration of explanted cells (trunk neural crest cells) is described. This method is inexpensive, gentle, and capable of distinguishing chemotaxis from both chemokinesis and other influences on migratory polarity such as those derived from cell-cell interactions within the primary trunk neural crest cell culture.

A method is described for efficient purification of twin-Strep-tagged fusion proteins and their specific complexes on modified streptavidin (Strep-Tactin) resin covalently cross-linked with Bis(sulfosuccinimidyl) suberate (BS3). The method has the advantages of fast speed, good target protein recovery and high purity, and is compatible with subsequent analysis by mass spectrometry.
