Calcium Green-5n

Calcium Green-5N is a fluorescent calcium indicator used to detect and quantify changes in free Ca2+ concentration, particularly in biochemical and cellular systems with high calcium levels. The dye contains calcium-binding sites that undergo a fluorescence increase when they bind Ca2+, allowing optical signals to report changes in ion availability; its relatively low affinity helps it respond within elevated calcium ranges. Researchers use Calcium Green-5N to monitor calcium flux, investigate ion transport and buffering, and characterize calcium-dependent processes in isolated organelles, solutions, and experimental preparations. These measurements support studies of cellular signaling, muscle function, and calcium homeostasis.

We present a method to isolate rapid (microsecond) calcium events from slower fluxes in living cells using laser scanning confocal microscopy. The method measures fluorescence intensity fluctuations of calcium indicators by recording line scans of several hundred pixels in a cell. Histogram analysis allows us to isolate the time scales of different calcium fluxes.
