High-concentration Formulation

High-concentration formulation is the design of solutions containing unusually large amounts of an active biomolecule or compound in a practical delivery volume, an important challenge in biochemistry and pharmaceutical development. Increasing concentration can improve dose delivery, but it may also raise viscosity, promote protein aggregation, reduce solubility, and intensify interactions among the active ingredient, excipients, and container surfaces; formulation scientists therefore optimize pH, ionic strength, stabilizers, and processing conditions. These formulations support compact injectable products, including biologic medicines administered by subcutaneous injection, while helping maintain potency, stability, manufacturability, and patient usability during storage and delivery.

Freeze-drying is often an easy and convenient way to obtain dry products of viable bacterial cells. An issue of the process is cell survival. We detail here a procedure to investigate how cell survival during freeze-drying is influenced by the properties of the formulation used.

Evaluating the Effect of a Synbiotic Formulation on Alzheimer’s Disease Pathology in Drosophila

Take a transgenic Drosophila. The brain of the Drosophila expresses the human amyloid peptide protein or APP and a cleaving enzyme. The enzyme cleaves APP, enabling the formation of beta-amyloid plaque, which leads toAlzheimer's disease or AD pathology in Drosophila.Feed the Drosophila AD model a diet containing a synbiotic formulation. The formulation contains a combination of three bioactive probiotic bacteria and a prebiotic.Inside the fly's gut, the prebiotic supports the growth and survival...

Source: Hemmerich, J., et al. Protocol for Optimization of Heterologous Protein Production Using Automated Microbioreactor Technology. J. Vis. Exp. (2017).This video demonstrates the use of an automated microbioreactor to monitor bacterial growth and fluorescent protein secretion in response to varying media formulations. A genetically modified bacterium expressing a secretory fluorescent protein is cultured under antibiotic selection and inoculated into nutrient-varied wells. Real-time optical...

Concentration Cells

A concentration cell is a type of a voltaic cell constructed by connecting two almost identical half-cells, both based on the same half-reaction and using the same electrode, differing only in the concentration of one redox species. A concentration cell's potential, therefore, is determined only by the concentration difference of the particular redox species. Consider the following voltaic cell: From the given information, the cell potential of this concentration cell can be calculated using...

Source: Smaa Koraym at Johns Hopkins University, MD, USA Concentration DependenceExpand In this experiment, you will combine varying concentrations of aqueous HCl and sodium thiosulfate to make solid sulfur, which rapidly gathers into opaque yellow particles at a certain sulfur concentration. Since the reaction solution starts clear and colorless, you can easily tell when it reaches that concentration. You'll use the same test tube and total volume each time, so it will take the same amount...
