Phospholipase C Beta

Phospholipase C beta (PLCβ) is a signaling enzyme that converts extracellular stimuli into intracellular biochemical responses, making it central to cell communication and biochemistry. Activated primarily by G protein-coupled receptors through Gq/11 proteins, PLCβ hydrolyzes the membrane phospholipid phosphatidylinositol 4,5-bisphosphate (PIP2) to produce inositol 1,4,5-trisphosphate (IP3) and diacylglycerol (DAG). IP3 releases calcium ions from the endoplasmic reticulum, while DAG activates protein kinase C, together regulating secretion, contraction, metabolism, and gene expression. Studying PLCβ helps clarify signal transduction pathways and supports research into neurological, cardiovascular, endocrine, and immune disorders linked to altered cellular signaling.

In this video, we demonstrate an assay to establish the presence of phospholipase in the crude venom extract from Anthopleura dowii—a marine sea anemone. The assay utilizes an agarose gel diffusion technique, in which the enzyme phospholipase diffuses and breaks down phospholipids incorporated in the solid media in a radial manner.

Herein we report a tailored HPLC purification protocol that yields high-purity amyloid beta 42 (Aβ42) and amyloid beta 40 (Aβ40) peptides, capable of oligomer formation. Amyloid beta is a highly aggregation prone, hydrophobic peptide implicated in Alzheimer's disease. The amyloidogenic nature of the peptide makes its purification a challenge.

To test the inhibitory effects of pharmacologic agents on phospholipase C (PLC) in different regions of the honeybee brain, we present a biochemical assay to measure PLC activity in those regions. This assay could be useful for comparing PLC activity among tissues, as well as among bees exhibiting different behaviors.
