Ammonium Sulfate Gradient

An ammonium sulfate gradient is a controlled range of ammonium sulfate concentrations used to separate proteins according to their solubility. As salt concentration increases, sulfate ions compete with proteins for water, reducing protein hydration and causing different molecules to precipitate at characteristic saturation levels; sequential addition, equilibration, and centrifugation can collect distinct fractions. In bioengineering, this salting-out method helps purify proteins from complex biological mixtures while maintaining biological activity. It can concentrate enzymes, antibodies, and recombinant proteins before chromatography, improve downstream recovery, and support process development by revealing protein solubility profiles.

In this video, we demonstrate a procedure for combined Alcian blue and silver staining of sulfated glycosaminoglycan oligosaccharide fragments. This combined staining technique significantly amplifies the bands of glycosaminoglycan polymers of different lengths in polyacrylamide gels.

Preparation and Characterization of SDF-1α-Chitosan-Dextran Sulfate Nanoparticles

The objective of this protocol is to incorporate SDF-1α, a stem cell homing factor, into dextran sulfate-chitosan nanoparticles. The resultant particles are measured for their size and zeta potential, as well as the content, activity, and in vitro release rate of SDF-1α from the nanoparticles.
