Atrial Ventricular Myocytes

Atrial and ventricular myocytes are specialized cardiac muscle cells that generate the electrical and mechanical activity required for coordinated heart function. Their membranes produce action potentials through precisely timed ion-channel currents; depolarization then triggers calcium entry and calcium release from the sarcoplasmic reticulum, enabling actin-myosin contraction. Atrial myocytes support blood transfer into the ventricles, whereas ventricular myocytes produce the force that ejects blood to the lungs and body. In bioengineering, these cells are used to build engineered cardiac tissues, model arrhythmias and inherited disease, evaluate drug effects, and develop electrophysiological platforms for regenerative medicine.

Simultaneous Isolation and Culture of Atrial Myocytes, Ventricular Myocytes, and Non-Myocytes from an Adult Mouse Heart

A method is described for the simultaneous isolation of myocytes and non-myocytes from both the atria and ventricles of a single adult mouse heart. This protocol results in consistent yields of highly viable cardiac myocytes and non-myocytes and details optimal cell-specific culture conditions for phenotyping and in vitro analysis.

Isolation and Kv Channel Recordings in Murine Atrial and Ventricular Cardiomyocytes

Kv channel dysfunction is associated with cardiac arrhythmias. In order to study the molecular mechanisms that lead to such arrhythmias we utilize a systematic protocol for isolation of atrial and ventricular cardiomyocytes from Kv channel ancillary subunit knockout mice. Isolated cardiomyocytes can then immediately be used for cellular electrophysiological studies, biochemical or immunofluorescence (IF) assays.

This protocol is used to isolate single atrial cardiomyocytes from the adult mouse heart using a chunk digestion approach. This approach is used to isolate right or left atrial myocytes that can be used to characterize atrial myocyte electrophysiology in patch-clamp studies.
