Exosome Cargo Analysis

Exosome cargo analysis is the study of the proteins, lipids, nucleic acids, and other molecular contents carried by exosomes, small extracellular vesicles that mediate communication between cells. Researchers isolate exosomes from biological fluids or cell cultures, verify their identity and purity, and characterize cargo using approaches such as sequencing, mass spectrometry, and targeted molecular assays. In bioengineering, these profiles help reveal how cells influence tissue behavior, disease processes, and immune responses through vesicle-mediated signaling. Cargo analysis also supports the design of engineered exosomes for biomarker development, therapeutic delivery, and regenerative medicine, while enabling assessment of cargo loading, stability, and functional effects.

Identification of Kinesin-1 Cargos Using Fluorescence Microscopy

Here, a protocol is presented to identify Kinesin-1 cargos. A motorless mutant of the Kinesin-1 heavy chain (KIF5B) aggregates in the cytoplasm and induces aggregation of its cargos. Both aggregates are detected under fluorescence microscopy. A similar strategy can be employed to identify cargos of other motor proteins.
