Intervertebral Disc Preparation

Intervertebral disc preparation is the controlled process of collecting, isolating, and conditioning spinal disc tissue for laboratory analysis, biomaterial development, or engineered tissue models. It typically involves separating disc regions such as the annulus fibrosus and nucleus pulposus while preserving their structure and composition and limiting mechanical or biochemical damage during handling. In bioengineering, careful preparation supports reproducible mechanical testing, cell-based studies, imaging, and evaluation of scaffolds or regenerative strategies. Standardized tissue handling also helps researchers compare healthy and degenerated discs and assess how engineered constructs may restore load-bearing function.

Intervertebral disc degeneration is a significant contributor to back pain and a leading cause of disability worldwide. Numerous animal models of intervertebral disc degeneration exist. We demonstrate an ovine model of intervertebral disc degeneration, utilizing a drill bit, which achieves a consistent disc injury and reproducible level of disc degeneration.
