Metabolite Fractionation

Metabolite fractionation is the systematic separation of small molecules from a biological sample into groups with related chemical or physical properties, enabling detailed analysis of cellular chemistry. The process typically combines selective extraction with techniques such as centrifugation, liquid-liquid partitioning, chromatography, or filtration, where differences in polarity, solubility, charge, size, or affinity determine how metabolites distribute between fractions. In bioengineering, fractionation helps isolate products from engineered cells or bioprocess streams, reduce sample complexity, and improve metabolite identification and quantification. These measurements support pathway analysis, strain optimization, process monitoring, and the development of more efficient systems for producing valuable biochemicals.

Mitochondria play central roles in the regulation of metabolism and homeostasis. Subtle changes in mitochondrial metabolism that affect organismal physiology could be difficult to detect in whole organism metabolomics studies. We describe here the procedures for the extraction and purification of mRNA and metabolites from Drosophila heads. We are applying these techniques to better understand the cellular perturbations underlying neuronal degeneration. These methodologies can be easily scaled and adapted for other "omic" projects.
