Neural Tissue Engineering

Neural tissue engineering is a bioengineering discipline that develops biological and synthetic strategies to restore, replace, or model damaged nervous tissue. It works by combining neural cells or progenitor cells with biomaterials, three-dimensional scaffolds, and biochemical or physical cues that support cell survival, guide neurite growth, and promote functional connectivity. These engineered systems can provide structured environments for studying neural development and disease, testing therapies, and designing regenerative approaches for injuries of the brain, spinal cord, and peripheral nerves. By linking material design with cellular behavior, the field seeks to improve neural repair while enabling more predictive in vitro models.

Here we demonstrate a novel silk-collagen-based 3D engineered model of neural tissue resembling brain-like architecture. The model can be used to study neuronal network assembly, axonal guidance, cell-cell interactions and electrical activity.

This manuscript details the fabrication of micro-tissue engineered neural networks: three-dimensional micron-sized constructs comprised of long aligned axonal tracts spanning aggregated neuronal population(s) encased in a tubular hydrogel. These living scaffolds can serve as functional relays to reconstruct or modulate neural circuitry or as biofidelic test-beds mimicking gray-white matter neuroanatomy.
