Phase Correlation Registration

Phase correlation registration is an image-alignment method that estimates the translation needed to superimpose two images, making it valuable for correcting motion and comparing biological structures. It uses the Fourier shift theorem: after transforming both images into the frequency domain, their normalized cross-power spectrum isolates phase differences caused by displacement, and an inverse Fourier transform produces a sharp peak at the relative shift. In bioengineering, the method supports microscopy, medical imaging, and time-lapse analysis by aligning frames despite changes in position or acquisition conditions. Its computational efficiency and reduced sensitivity to intensity variations help quantify structural changes, track specimens, and improve downstream image analysis.

A method is described whereby quantum dot (QD) nanoparticles can be used for correlative immunocytochemical studies of epoxy embedded human pathology tissue. We employ commercial antibody fragment conjugated QDs that are visualized by widefield fluorescence light microscopy and transmission electron microscopy.
