Prp Sirna

PrP siRNA is a gene-silencing approach that reduces production of the cellular prion protein, PrP, by targeting its messenger RNA. After entering cells, the small interfering RNA is loaded into the RNA-induced silencing complex, which uses sequence complementarity to recognize and cleave PRNP mRNA, limiting translation into PrP. In bioengineering, PrP siRNA supports functional studies of prion protein biology and enables evaluation of RNA delivery systems, including engineered nanoparticles and other carriers. By linking sequence-specific regulation with controlled delivery, this approach can help investigate prion-associated mechanisms and inform the development of potential intervention strategies.

Implant-associated infection is a significant clinical complication. This study describes an approach using platelet-rich plasma (PRP) to prevent implant-associated infections, presents the protocol for preparing PRP with constant platelet concentration, and reports the newly identified antimicrobial properties of PRP and related protocols for examining such antimicrobial properties in vitro.

Targeted siRNA Delivery to Neurons for Prion Protein Silencing Using Liposomal Complexes

Source: Bender, H. R., et.al. Delivery of Therapeutic siRNA to the CNS Using Cationic and Anionic Liposomes. J. Vis. Exp. (2016)This video demonstrates the targeted delivery of prion protein (PrP) siRNA to neurons using liposomal siRNA-peptide complexes (LSPCs). Cationic liposomes electrostatically bind PrP-targeting siRNA, which is further stabilized by the RVG-9r peptide for receptor-mediated transport across the blood-brain barrier. Once inside the brain, LSPCs undergo endocytosis in...

siRNA - Small Interfering RNAs

2020

Small interfering RNAs, or siRNAs, are short regulatory RNA molecules that can silence genes post-transcriptionally, as well as the transcriptional level in some cases. siRNAs are important for protecting cells against viral infections and silencing transposable genetic elements. In the cytoplasm, siRNA is processed from a double-stranded RNA, which comes from either endogenous DNA transcription or exogenous sources like a virus. This double-stranded RNA is then cleaved by the ATP-dependent...
