Stop-flow Technique

The stop-flow technique is a rapid-mixing method for measuring the time-dependent behavior of chemical and biological reactions. It combines reactants in a flow system, then abruptly stops the motion of the solution so instruments can monitor changes in absorbance, fluorescence, or other signals over short time intervals. In bioengineering, this approach helps quantify enzyme kinetics, ligand binding, protein conformational changes, and biomolecular interactions that occur too quickly for conventional sampling. By resolving reaction intermediates and rate constants, stop-flow measurements support the design of biosensors, therapeutic proteins, diagnostic assays, and engineered biochemical systems.

The topical anesthetic lidocaine was atomized using a high oxygen flow through the working channel of a flexible intubating endoscope to achieve topical airway anesthesia for awake endotracheal intubation. We prefer this modified spray-as-you-go technique for endoscopic intubation to classical bolus application because of higher patient satisfaction and better compliance.

Intracellular Ca2+ dynamics are very important in sperm physiology and Ca2+-sensitive fluorescent dyes constitute a versatile tool to study them. Population experiments (fluorometry and stopped flow fluorometry) and single cell experiments (flow cytometry and single cell imaging) are used to track spatio-temporal [Ca2+] changes in human sperm cells.

In this video, we present a photoacoustic flow cytometric detection of ovarian cancer cells bearing nanoparticles in a circulating in vitro flow chamber. This non-invasive technique successfully identifies circulating tumor cells in a flow system utilizing a contrast agent.
