Adipose Tissue Isolation

Adipose tissue isolation is the laboratory process of separating fat tissue from surrounding materials to obtain intact tissue or its cellular components for research and engineering. Typically, collected tissue is minced and processed through enzymatic digestion and mechanical disruption, followed by filtration and centrifugation to separate adipocytes, stromal vascular cells, and extracellular matrix fractions. In bioengineering, isolated adipose tissue provides cells and matrix components for studying tissue structure, developing engineered adipose constructs, and investigating regenerative strategies. The resulting preparations support analyses of cell behavior, scaffold interactions, and tissue remodeling under controlled experimental conditions.

Adipose tissue (AT) is a site of intense immune cell activation and interaction. Almost all cells of the immune system are present in AT and their ratios are altered by obesity. Proper isolation, quantification, and characterization of AT immune cell populations are critical for understanding their role in immunometabolic disease.

This video demonstrates a technique for isolating and culturing fluorescently labeled neural crest-derived adipose-derived stem cells (ADSCs) from murine periaortic arch adipose tissue. Following isolation, the NCADSCs are detected, sorted using fluorescence-activated cell sorting, and subsequently cultured on a plate, promoting cell proliferation.

Isolation and Differentiation of Adipose-Derived Stem Cells from Porcine Subcutaneous Adipose Tissues

This protocol describes the isolation of pig adipose-derived stem cells (pADSC) from subcutaneous adipose tissues with examination of multipotency. The multipotent pADSC are used to delineate processes of adipocyte differentiation and study transdifferentiation into multiple cell lineages of mesodermal mesenchyme or further lineages of ectoderm and endoderm for regenerative studies.

We present a protocol to isolate adipose tissue-derived microvascular fragments that represent promising vascularization units. They can be rapidly isolated, do not require in vitro processing and, thus, may be used for one-step prevascularization in different fields of tissue engineering.
