Vascular Endothelial Cells

Vascular endothelial cells are specialized cells that form the inner lining of blood vessels, creating a dynamic interface between circulating blood and surrounding tissues. They regulate vascular permeability, blood flow, inflammation, and vessel formation by responding to biochemical signals and mechanical forces such as fluid shear stress; their junctions and signaling pathways adjust barrier function and promote angiogenesis when needed. In bioengineering, vascular endothelial cells help generate blood vessel models, assess biomaterials, and vascularize engineered tissues. Their behavior provides essential benchmarks for studying cardiovascular disease, drug responses, implant compatibility, and strategies for building functional tissue-engineered organs.

Assessing Mitochondrial Function in Cerebral Vascular Endothelial Cells

This video demonstrates the assessment of the mitochondrial functioning of cerebral vascular endothelial (CVE) cells using an extracellular flux assay. CVE cells are plated in an extracellular flux cell culture plate. Extracellular flux sensors are hydrated before loading and calibration into the bioanalyzer. The plate containing cells is then loaded for analysis.

This work demonstrates a novel approach to assess the proliferation, differentiation, and vessel-forming potential of vascular endothelial stem cells (VESCs) through mammary fat pad transplantation followed by whole-mount tissue preparation for microscopic observation. A lineage tracing strategy to investigate the behavior of VESCs in vivo is also presented.

A series of methods is described, including the isolation of low-density lipoprotein (LDL) from human plasma, differentiation of HL-60 cells into neutrophil-like cells, preparation of neutrophil extracellular traps (NETs) in the presence of LDL, and stimulation of human aortic endothelial cells with a mixture of NETs and LDL.

A Simple Bioassay for the Evaluation of Vascular Endothelial Growth Factors

We describe a simple cell-based bioassay for detecting, quantifying and monitoring the activity of members of the vascular endothelial growth factor family of ligands. The assay uses chimeric receptors expressed in a factor-dependent cell line to provide a semi-quantitative or quantitative assessment of receptor binding and cross-linking by the ligand.
