Antibody Probing Protocol

An antibody probing protocol is a laboratory method for detecting and characterizing a specific protein or other antigen in a biological sample, supporting research in molecular and cellular biology. The procedure typically exposes a membrane, tissue section, or cell preparation to a primary antibody that binds the target through selective antigen recognition, followed by a labeled secondary antibody that produces a fluorescent or enzymatic signal. Researchers use antibody probing in techniques such as Western blotting, immunohistochemistry, and immunofluorescence to measure protein abundance, determine cellular location, and assess changes in signaling or disease-related pathways. Careful blocking and washing steps improve specificity and reduce background.

The development of new ultrasound (US) probes based on Capacitive Micromachined Ultrasonic Transducer (CMUT) technology requires an early realistic assessment of imaging capabilities. We describe a repeatable experimental protocol for US image acquisition and comparison with magnetic resonance images, using an ex vivo bovine brain as an imaging target.

Closed-loop protocols are becoming increasingly widespread in modern day electrophysiology. We present a simple, versatile and inexpensive way to perform complex electrophysiological protocols in cortical pyramidal neurons in vitro, using a desktop computer and a digital acquisition board.

Scalable High Throughput Selection From Phage-displayed Synthetic Antibody Libraries

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.
