Dna Denaturation Neutralization

DNA denaturation and neutralization are complementary molecular biology procedures that separate double-stranded DNA and then restore conditions that support strand pairing. Denaturation commonly uses heat or alkaline pH to disrupt hydrogen bonding between complementary bases, while neutralization returns the solution toward a suitable pH, allowing complementary strands to reanneal. In biological techniques, controlled denaturation exposes nucleotide sequences for hybridization, amplification, sequencing, and probe-based detection. During alkaline plasmid preparation, neutralization also helps precipitate proteins and chromosomal DNA while preserving soluble plasmid DNA, making the step important for selective purification and downstream analysis.

In this video, we demonstrate an in vitro colorimetric assay to detect the presence of neutralizing antibodies against alkaline phosphatase-expressing recombinant adeno-associated virus serotype 6 (rAAV6) vectors in sheep serum samples. The assay utilizes the reaction between the AAV encoding a human placental alkaline phosphatase gene and its substrate to generate an insoluble purple product.

Denaturing urea polyacrylamide gel electrophoresis is used to separate single-stranded DNA or RNA up to a limit of 500 nucleotides. Urea in combination with heat denatures samples and unstructured single strands migrate within the gel matrix according to their molecular weight.

This protocol is designed to investigate the extent of DNA damage occurring during DNA replication.
