Scintillation Counter Measurement

Scintillation counter measurement is a radioactivity-detection method that quantifies ionizing radiation by converting radioactive decay into flashes of light, making it valuable for sensitive tracer analysis. In liquid scintillation counting, a radiolabeled sample is mixed with a scintillator cocktail; emitted particles transfer energy to the cocktail, which produces photons detected by photomultiplier tubes and reported as counts or disintegrations per minute. In biological techniques, researchers use the method to measure radiolabeled DNA, RNA, proteins, metabolites, and cellular uptake. Its high sensitivity supports studies of metabolism, biosynthesis, transport, and ligand binding, while calibration and quench correction improve quantitative accuracy.

Radiolabeling and Quantification of Cellular Levels of Phosphoinositides by High Performance Liquid Chromatography-coupled Flow Scintillation

Phosphoinositides are signaling lipids whose relative abundance rapidly changes in response to various stimuli. This article describes a method to measure the abundance of phosphoinositides by metabolically labeling cells with 3H-myo-inositol, followed by extraction and deacylation. Extracted glycero-inositides are then separated by high-performance liquid chromatography and quantified by flow scintillation.
