Perchloric Acid Quench

Perchloric acid quench is a sample-preparation method that rapidly stops biological reactions and preserves the chemical state of metabolites for analysis. Adding perchloric acid sharply lowers pH, denatures and precipitates proteins, and inactivates enzymes, separating soluble small molecules from macromolecular material; subsequent neutralization and centrifugation can produce a clarified extract. In biological techniques, this approach supports measurements of cellular metabolites, nucleotides, and other acid-stable compounds by limiting changes that occur after sampling. Because perchloric acid is highly corrosive and a strong oxidizer, quenching requires controlled handling, compatible equipment, and appropriate safety procedures.

Quorum-quenching enzymes are anti-virulent and anti-bacterial options that can mitigate pathogenesis without risk of incurring resistance, by preventing the expression of virulence factors and genes associated with antibiotic resistance and biofilm formation. In this study, we report a method that demonstrates the efficacy of quorum-quenching enzymes in bacterial biofilm disruption.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.

This protocol details an in vitro assay for measuring cellular lipid uptake in endothelial cells upon stimulation with BODIPY-C12 and BODIPY-C16 fluorescent analogs of long-chain and very long-chain saturated fatty acids. This method is efficient and adaptable to other cell types, offering a useful approach for studying lipid metabolism.
