Actin Filament Preparation

Actin filament preparation is a laboratory method for generating organized filaments of actin, a cytoskeletal protein that supports cell shape, movement, and intracellular organization. The process typically begins with soluble globular actin, or G-actin, which polymerizes into filamentous F-actin when suitable ionic conditions, cofactors, and energy from ATP promote nucleation and elongation; the resulting filaments can then be stabilized or labeled for analysis. Prepared actin filaments provide a controlled system for studying cytoskeletal assembly, motor protein activity, membrane interactions, and force generation, while also supporting microscopy, biochemical assays, and investigations of cell motility.

Filamentous actin (F-actin) plays an important role in spinogenesis, synaptic plasticity, and synaptic stability. Quantification of F-actin puncta is therefore a useful tool to study the integrity of synaptic structures.
