Protein Binding Analysis

Protein binding analysis is the study of how proteins interact with other molecules, including ligands, nucleic acids, lipids, and small-molecule drugs, and why these interactions matter in biology. The analysis measures binding strength and behavior by examining how molecules associate and dissociate, often under controlled concentrations and conditions to determine affinity, specificity, and binding-site characteristics. These measurements help explain protein function, signaling, transport, and regulation while supporting the characterization of enzymes, receptors, antibodies, and therapeutic compounds. In research and drug development, protein binding analysis can reveal molecular mechanisms, guide compound optimization, and predict how changes in molecular structure may alter biological activity.

Here, we present human cell culture protocols to analyze translation initiation factors that bind the 5' cap of mRNA during physiological oxygen conditions. This method utilizes an Agarose-linked m7GTP cap analog and is suitable to investigate cap-binding factors and their interacting partners.

Competition Binding Assay to Study Competing GTPase-Binding Protein Partners

2025

This video demonstrates a competition assay to study GTPase-binding protein partners. Utilizing nucleotide-bound GTPase protein immobilized on magnetic beads, the competitive binding between two interacting protein partners for the same binding site on the GTPase can be studied to assess the binding affinities of the protein partners.
