Acute Myeloid Leukemia

Acute myeloid leukemia (AML) is an aggressive cancer of the blood and bone marrow in which abnormal myeloid precursor cells multiply rapidly and interfere with normal blood formation. Genetic changes disrupt pathways that control cell differentiation and proliferation, causing immature myeloid blasts to accumulate in the marrow and reduce production of healthy red blood cells, platelets, and white blood cells. This can lead to anemia, bleeding, infection, and other complications. Studying AML supports the classification of disease subtypes, development of diagnostic tests, and evaluation of targeted therapies, chemotherapy, and stem cell transplantation, while also providing insight into cancer biology and blood-cell development.

This new method is effective in the serial transplantation of AML cells and can serve as an alternative for those who may experience difficulties and inconsistencies with intravenous injection in mice.

Intracellular Phosphoflow Cytometry of Acute Myeloid Leukemia Patient-Derived Xenotransplants

Here, a phosphoflow cytometry-based method is described to analyze signaling downstream of the mTORC1, JAK/STAT5, and MAPK pathways in acute human myeloid leukemia cells xenografted into mice and obtained from bone marrow aspirates. p-STAT5, p-4EBP1, p-RPS6, and p-ERK1/2 levels are simultaneously measured using a next-generation spectral flow cytometer with high sensitivity.
