Adipocyte Aggregate Formation

Adipocyte aggregate formation is the organization of adipocytes into multicellular, three-dimensional clusters that model aspects of adipose tissue architecture. The process relies on cell-cell adhesion and interactions with the extracellular matrix, often under conditions that limit attachment to a flat culture surface and promote close cellular contact. As aggregates develop, their tissue-like microenvironment can affect signaling, lipid storage, nutrient exchange, and adipocyte differentiation. In biology research, this model supports investigation of adipose tissue organization, metabolic function, obesity-related mechanisms, and cellular responses to candidate therapeutics, while complementing conventional two-dimensional adipocyte cultures.

Protein Aggregate Formation Assay: A Method to Detect and Quantify Protein Aggregation in Cultured Cells upon Induction by Proteasome Inhibitor

In this video, we demonstrate a cell-based protein aggregation assay using proteasome inhibitors, which block proteasome activity, preventing misfolded, mutant proteins, fused to a fluorescent label, from undergoing ubiquitin-dependent proteasomal degradation, leading to their accumulation within the cell cytoplasm. The protein aggregates are then visualized and quantified by fluorescence microscopy.

Adipose tissue (AT) can influence whole body homeostasis, therefore understanding the molecular mechanisms of adipocyte differentiation and function is of importance. We provide a protocol for gaining new insights into these processes by analyzing adipocyte homeostasis, differentiation and hypoxia exposure as a model for induced adipocyte apoptosis.

This video demonstrates the differentiation of bone marrow stromal cells (BMSCs) into adipocytes. In the two-step process, in the presence of specific growth factors and hormones, undifferentiated BMSCs first differentiate into preadipocytes. The second step involving treatment with glucose and insulin leads to the formation of mature adipocytes with multiple lipid droplets.

Mature adipocytes may represent an abundant source of stem cells through dedifferentiation, which leads to a homogenous population of fibroblast-like cells. Collagenase digestion is used to isolate mature adipocytes from human fat. The goal of our protocol is to obtain multipotent, dedifferentiated fat cells from human mature adipocytes.
