Adipose Turnover

Adipose turnover is the ongoing renewal and remodeling of fat tissue, a biological process that influences energy balance, body composition, and metabolic health. It reflects coordinated changes in adipocyte number and size, including precursor-cell differentiation, triglyceride storage and mobilization, and removal or replacement of mature cells. Hormonal and nutritional signals regulate lipolysis and lipid uptake, while inflammation and tissue remodeling can alter how adipose tissue expands or contracts. Studying adipose turnover helps explain obesity, insulin resistance, and metabolic adaptation, supporting research into therapies that promote healthy fat storage and systemic metabolic function.

Isolating Adipose-Derived Stem Cells from Murine Periaortic Adipose Tissue

This video demonstrates a technique for isolating and culturing fluorescently labeled neural crest-derived adipose-derived stem cells (ADSCs) from murine periaortic arch adipose tissue. Following isolation, the NCADSCs are detected, sorted using fluorescence-activated cell sorting, and subsequently cultured on a plate, promoting cell proliferation.

This protocol describes the isolation of pig adipose-derived stem cells (pADSC) from subcutaneous adipose tissues with examination of multipotency. The multipotent pADSC are used to delineate processes of adipocyte differentiation and study transdifferentiation into multiple cell lineages of mesodermal mesenchyme or further lineages of ectoderm and endoderm for regenerative studies.

Adipose tissue (AT) is a site of intense immune cell activation and interaction. Almost all cells of the immune system are present in AT and their ratios are altered by obesity. Proper isolation, quantification, and characterization of AT immune cell populations are critical for understanding their role in immunometabolic disease.

Steady-state, Pre-steady-state, and Single-turnover Kinetic Measurement for DNA Glycosylase Activity

Time courses for the glycosylase activity of 8-oxoguanine DNA glycosylase are biphasic exhibiting a burst of product formation and a linear steady-state phase. Utilizing quench-flow techniques, the burst and the steady-state rates can be measured, which correspond to excision of 8-oxoguanine and release of the glycosylase from the product DNA, respectively.
