Agilent 2100 Bioanalyzer

The Agilent 2100 Bioanalyzer is a microfluidic laboratory instrument that rapidly evaluates the size, concentration, and integrity of DNA, RNA, and some protein samples, making it useful for quality control in biological research. It works by moving samples through disposable Lab-on-a-Chip channels, where electrophoretic separation resolves molecules according to size; fluorescent labeling and detection then generate electropherograms and numerical measurements. Researchers use these data to assess RNA integrity before gene-expression studies or sequencing, verify DNA libraries and PCR products, and examine protein samples. By standardizing sample analysis and reducing hands-on processing, the system helps identify poor-quality material early and supports reproducible experimental decisions.

In vitro Transcription and Capping of Gaussia Luciferase mRNA Followed by HeLa Cell Transfection

This method describes high yield in vitro synthesis of both capped and uncapped mRNA from a linearized plasmid containing the Gaussia luciferase (GLuc) gene. The RNA is purified and a fraction of the uncapped RNA is enzymatically capped using the Vaccinia virus capping enzyme. In the final step, the mRNA is transfected into HeLa cells and cell culture supernatants are assayed for luciferase activity.

Alcohol misuse impairs alveolar macrophage (AM) immunity due to suppressed mitochondrial respiration and bioenergetics. We recently demonstrated that ethanol (EtOH) exposure increases glutamine dependency for mitochondrial respiration in AMs. Herein, methods are provided to determine the usage of glutamine for mitochondrial respiration in EtOH-treated AMs using an extracellular flux bioanalyzer.

Isolation and Purification of Drosophila Peripheral Neurons by Magnetic Bead Sorting

In this video-article we present a method for the isolation and purification of Drosophila peripheral neurons using a fast magnetic bead assisted cell sorting strategy. RNA obtained from the isolated cells can be readily used for downstream applications including microarray analyses.
