Alzheimer's Disease Mouse Model

An Alzheimer’s disease mouse model is a laboratory mouse engineered or treated to reproduce selected features of Alzheimer’s disease, providing a controlled system for studying its biology. Many models express human disease-associated genes, such as mutant amyloid precursor protein or presenilin, which can increase amyloid-beta production and promote plaque formation; other models examine tau pathology, neuroinflammation, or cognitive impairment. Researchers use these models to investigate disease mechanisms, evaluate biomarkers, and test potential drugs or preventive strategies. Because no single model reproduces the full human condition, comparing complementary models helps clarify which findings may translate to patients.

One of the pathological characteristics of AD is the formation of Amyloid β protein positive neuritic plaques. In this protocol we describe two methods to detect neuritic plaques in transgenic AD model mice: immunohistochemical detection using the ABC and DAB method and fluorescent detection using thioflavin S staining method.
