Aphid Vector

An aphid vector is an aphid that acquires and transmits infectious agents, especially plant viruses, between host plants, making it important in plant biology, agriculture, and disease ecology. During feeding, the aphid inserts its stylets into plant tissues; viruses may adhere to the mouthparts and move rapidly between plants during brief probes, or circulate through the gut and hemolymph before reaching the salivary glands during sustained feeding. Studying aphid vectors clarifies how plant diseases spread, how host and vector interactions shape epidemics, and how monitoring, resistant crops, and integrated pest management can reduce transmission.

Aphids are effective transmitters of plant viruses. Aphid microinjection of virus, the procedure we will show you today, is a technique allowing researchers to inject virus directly into the hemocoel of the aphid, bypassing the gut, one of the 2 major barriers for virus transmission in a circulative manner. The same technique is also used to inject dsRNA for RNAi.

A protocol of whole-mount immunostaining on aphid embryos is presented, in which critical conditions for decreasing background staining and increasing tissue permeability are addressed. These conditions are specifically developed for effective detection of protein expression in the embryonic tissues of aphids.

We describe the production strategy of integrase-deficient lentiviral vectors (IDLVs) as vehicles for delivering CRISPR/Cas9 to cells. With an ability to mediate quick and robust gene editing in cells, IDLVs present a safer and equally effective vector platform for gene delivery compared to integrase-competent vectors.
