Automated Protocol

An automated protocol is a programmed, repeatable sequence of laboratory steps executed by instruments or robotic systems, reducing manual handling in biological research. Software directs operations such as liquid transfer, reagent dispensing, mixing, incubation, washing, and measurement according to defined parameters, while sensors or programmed checks help maintain timing and volume consistency. Automated protocols support high-throughput experiments including sample preparation, nucleic acid extraction, cell-based assays, and screening workflows. By improving reproducibility, processing many samples efficiently, and limiting operator-dependent variation, they help laboratories standardize procedures and generate reliable data for research, diagnostics, and biotechnology.

We present a protocol on how to utilize high-throughput cryo-electron tomography to determine high resolution in situ structures of molecular machines. The protocol permits large amounts of data to be processed, avoids common bottlenecks and reduces resource downtime, allowing the user to focus on important biological questions.

This manuscript describes a generic approach for tailor-made design of microbial cultivation media. This is enabled by an iterative workflow combining Kriging-based experimental design and microbioreactor technology for sufficient cultivation throughput, which is supported by lab robotics to increase reliability and speed in liquid handling media preparation.
