Bacillales Order

Bacillales is an order of bacteria within the class Bacilli, encompassing diverse Gram-positive lineages with major roles in ecology, biotechnology, and human health. Members commonly reproduce by binary fission, while many Bacillus-related species respond to nutrient limitation by forming durable endospores that resist heat, drying, and chemical stress; other lineages, including Staphylococcus and Listeria, remain non-spore-forming. Studying Bacillales supports bacterial classification and evolutionary research while informing investigations of soil nutrient cycling, industrial enzyme and antibiotic production, food fermentation, and infectious disease. Their contrasting lifestyles also provide useful models for bacterial survival, adaptation, and host interaction.

Lentiviral Mediated Gene Silencing in Human Pseudoislet Prepared in Low Attachment Plates

A protocol to create gene modified human pseudoislets from dispersed human islet cells that are transduced by lentivirus carrying short hairpin RNA (shRNA) is presented. This protocol utilizes readily available enzyme and culture vessels, can be performed easily, and produces genetically modified human pseudoislets suitable for functional and morphological studies.
