Bacterial Motility

Bacterial motility is the ability of bacteria to move through liquid environments or across surfaces, shaping how they find nutrients, avoid harmful conditions, and colonize new habitats. Many bacteria swim using flagella, whose rotary motors convert the proton motive force across the cell membrane into movement, while others spread through surface-associated mechanisms such as type IV pilus retraction. Chemotaxis receptors detect chemical gradients and adjust flagellar activity, directing cells toward favorable conditions or away from repellents. Studying bacterial motility helps explain infection, biofilm formation, environmental adaptation, and microbial behavior, while supporting research on antimicrobial strategies and engineered biological systems.

Swarming motility is influenced by physical and environmental factors. We describe a two-phase protocol and guidelines to circumvent the challenges commonly associated with swarm assay preparation and data collection. A macroscopic imaging technique is employed to obtain detailed information on swarm behavior that is not provided by current analysis techniques.
