Barnyardgrass

Barnyardgrass (Echinochloa crus-galli) is a widespread annual grass and important agricultural weed, making it relevant to the study of plant growth, adaptation, and competition. It reproduces primarily through abundant seeds and uses C4 photosynthesis, a carbon-fixation pathway that improves carbon gain under high light, warmth, and limited water, helping seedlings grow rapidly in disturbed fields. In rice and other crops, barnyardgrass competes for light, nutrients, and water, reducing productivity and complicating field management. Its variable responses to herbicides also support research on plant evolution, resistance, and sustainable weed-control strategies.

We have developed a methodology for performing crosses in Setaria viridis (S. viridis). The method involves pruning the panicle prior to a hot water treatment to kill viable pollen. Crosses are performed following a well-controlled growth regime and typically result in the recovery of 1 to 7 cross-pollinated seed/s per panicle.

Excavation of plant roots from the field as well as processing of samples into endosphere, rhizosphere, and soil are described in detail, including DNA extraction and data analysis methods. This paper is designed to enable other laboratories to use these techniques for the study of soil, endosphere, and rhizosphere microbiomes.

We provide a cost-effective and rapid molecular genotyping protocol that employs variety-specific PCR primers that target DNA sequence differences within the chloroplast trnL-F spacer region to differentiate between varieties of Imperata cylindrica (cogongrass) that cannot be distinguished by morphology alone. These varieties include the federally listed noxious weed, cogongrass and closely-related, wide-spread ornamental variety, I. cylindrica var. koenigii (Japanese blood grass).

Linking Predation Risk, Herbivore Physiological Stress and Microbial Decomposition of Plant Litter

We present methods to evaluate how predation risk can alter the chemical quality of herbivore prey by inducing dietary changes to meet demands of heightened stress, and how the decomposition of carcasses from these stressed herbivores slows subsequent plant litter decomposition by soil microbes.
