Bmdm Cells

BMDM cells, or bone marrow-derived macrophages, are immune cells generated from bone marrow progenitors in culture and used to model macrophage biology under controlled laboratory conditions. Bone marrow cells differentiate into macrophages when cultured with macrophage colony-stimulating factor, then respond to environmental signals through pattern-recognition receptors and alter cytokine production, phagocytosis, and gene expression. Researchers use BMDM cells to study innate immunity, inflammation, host-pathogen interactions, tissue repair, and responses to drugs or genetic perturbations. Because they retain key macrophage functions in a tractable culture system, BMDMs help connect cellular mechanisms with disease-relevant experimental outcomes.

Here we describe a method for bacterial RNA isolation from Listeria monocytogenes bacteria growing inside murine macrophages. This technique can be used with other intracellular pathogens and mammalian host cells.

Isolation of Intracellular Pathogenic Bacteria from Infected Host Cells

Source: Sigal, N., et al. RNA Purification from Intracellularly Grown Listeria monocytogenes in Macrophage Cells. J. Vis. Exp. (2016).This video demonstrates the isolation of internalized pathogenic bacteria from infected bone marrow-derived macrophages (BMDMs). Bacteria are allowed to attach to host cells, and unattached bacteria are washed away. After internalization, an antibiotic is added to eliminate non-internalized bacteria. The host cells are lysed, centrifuged, and the supernatant is...

Investigation of Macrophage Polarization Using Bone Marrow Derived Macrophages

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.
