Brown Fat Knockout

Brown fat knockout is an experimental strategy that removes brown adipose tissue or disables genes required for its function, allowing researchers to determine how brown fat contributes to whole-body physiology. In a common approach, genetic deletion disrupts thermogenic components such as uncoupling protein 1 (UCP1), which normally dissipates the mitochondrial proton gradient as heat instead of producing ATP. These models help clarify brown fat’s roles in cold-induced thermogenesis, energy expenditure, glucose and lipid metabolism, and body-weight regulation. In biology and metabolic research, knockout studies can reveal potential targets for treating obesity, diabetes, and impaired temperature regulation while distinguishing brown-fat-specific effects from broader systemic responses.

In this protocol, we describe the technical procedures to generate brown adipose tissue (BAT)-specific knockout mice leveraging a combined Cre-LoxP, CRISPR-Cas9, and adeno-associated virus (AAV) single-guide RNA (sgRNA) system. The described steps include the design of the sgRNAs, the preparation of the AAV-sgRNA particles, and the microinjection of AAV into the BAT lobes.

A method of functional imaging of mouse brown adipose tissue (BAT) is described in which cold-stimulated uptake of 18F-Fluorodeoxyglucose (FDG) in BAT is non-invasively assessed with a standardized micro-PET/CT protocol. This method is robust and sensitive to detect differences in BAT activities in mouse models.

Transplantation Into the Mouse Ovarian Fat Pad

We describe an ovarian fad pad transplantation assay suitable for studies of normal and transformed epithelia of the female reproductive tract. The mouse fat pad allows transplantation of large tissue fragments, is easily accessible for surgery and imaging, and provides the most favorable native environment for tissues of Müllerian origin.
