Epidermal Growth Factor Removal

Epidermal growth factor removal is the deliberate depletion or withdrawal of epidermal growth factor (EGF) from a biological system, commonly cell culture, to examine how EGF-dependent signaling affects cell behavior. Replacing EGF-containing medium with EGF-free conditions reduces activation of the epidermal growth factor receptor and downstream pathways that regulate proliferation, survival, and cellular state. This approach helps distinguish EGF-specific effects from other growth-factor responses and can promote measurable changes in cell growth or differentiation. In biology, EGF removal supports studies of stem cells, organoids, tissue models, and receptor signaling, providing a controlled way to investigate growth-factor requirements and cellular adaptation.

This protocol describes the labeling of epidermal growth factor receptor (EGFR) on COS7 fibroblast cells, and subsequent correlative light- and electron microscopy of whole cells in hydrated state. The label contained fluorescent quantum dots. The protocol can be used to study the stoichiometry of EGFR at the single molecule level.

Studies of neuronal morphogenesis using Drosophila larval dendritic arborization (da) neurons benefit from in situ visualization of neuronal and epidermal proteins by immunofluorescence. We describe a procedure that improves immunofluorescence analysis of da neurons and surrounding epidermal cells by removing muscle tissue from the larval body wall.

We describe a simple cell-based bioassay for detecting, quantifying and monitoring the activity of members of the vascular endothelial growth factor family of ligands. The assay uses chimeric receptors expressed in a factor-dependent cell line to provide a semi-quantitative or quantitative assessment of receptor binding and cross-linking by the ligand.
