Cell Density Control

Cell density control is the measurement and adjustment of the number of cells occupying a defined volume or surface area, a key factor in maintaining reproducible biological experiments. Researchers regulate density through cell counting, dilution, controlled seeding, and monitoring confluence, while cells respond to available nutrients, growth factors, space, and cell-cell signaling. In cell culture and tissue studies, consistent density supports reliable growth, differentiation, viability, and gene-expression measurements; changing density can also model conditions such as proliferation, contact inhibition, or crowding. Careful control therefore improves experimental comparability and helps optimize applications in disease research, drug testing, and tissue engineering.

In Vivo Assay to Correlate Bacterial Bioluminescence with Cell Density

2025

Source: Brodl, E., et al. In Situ Measurement and Correlation of Cell Density and Light Emission of Bioluminescent Bacteria. J. Vis. Exp. (2018).This video demonstrates the measurement and correlation of bioluminescence with cell density in transformed bacteria. A single bacterial colony is used to initiate a liquid culture. The culture is placed in a well at the desired cell concentration. An antibiotic and inducer are added to express the gene required for bacterial bioluminescence. Readings...

Density Gradient Ultracentrifugation

2023

Density gradient ultracentrifugation is a common technique used to isolate and purify biomolecules and cell structures. This technique exploits the fact that, in suspension, particles that are more dense than the solvent will sediment, while those that are less dense will float. A high-speed ultracentrifuge is used to accelerate this process in order to separate biomolecules within a density gradient, which can be established by layering liquids of decreasing density in a centrifuge tube. The...

Studying the Protein Quality Control System of D. discoideum Using Temperature-controlled Live Cell Imaging

2016

The social amoebae Dictyostelium discoideum has recently been established as a system to study protein misfolding and proteostasis. Here, we describe a new imaging-based methodology to study temperature-induced protein aggregation and the cellular stress response in D. discoideum.

Generating an Ultra-Low-Density Neuronal Culture Using a High-Density Neuronal Feeder Layer

2025

This video demonstrates the method for culturing ultra-low-density neurons in the presence of a high-density neuronal feeder layer. It establishes a co-culture of varying-density neurons, ensuring close physical proximity. The growth factors secreted by high-density neurons help neuronal survival and growth, maintaining ultra-low-density neurons for a longer time period.

Peritoneal Low-density Neutrophil Isolation: A Technique to Obtain Low-density Neutrophils from Peritoneal Lavage Fluid Using Magnetic Activated Cell Sorting

2023

In this video, we describe the procedure for isolating low-density neutrophils, or LDNs, from the peritoneal lavage fluid of a postoperative human patient. The LDNs thus obtained can be used in further downstream experiments to understand their role in tumor growth and progression.
