Cell Pellet Controls

Cell pellet controls are standardized cell samples collected and processed as compact pellets to provide reference points for evaluating biological assays and sample handling. During preparation, centrifugation sediments cells from suspension, while washing, removing the supernatant, and resuspending or retaining the pellet establish consistent conditions for comparison; parallel untreated or known-condition samples can serve as controls. In biology, these controls help distinguish true experimental signals from background caused by cell recovery, lysis, processing, or loading differences. They support reliable normalization and interpretation in applications such as protein analysis, nucleic acid measurements, microscopy, and other workflows that begin with harvested cells.

Liquid-grown Streptomyces cultures are characterized by mycelial pellets that are heterogeneous in size. We here describe a method to analyze and sort such pellets in a high-throughput manner. These pellets can be used for further analyses, which will provide leads to understand and control growth heterogeneity.
