Rodent Heart Models

Rodent heart models are experimental systems that use mice or rats to investigate cardiac structure, function, disease, and treatment responses in a living organism or isolated tissue. They work by reproducing cardiovascular conditions through genetic modification, surgical intervention, pharmacological exposure, or controlled ex vivo perfusion, allowing researchers to measure contractility, electrical activity, hemodynamics, and tissue remodeling. These models support studies of myocardial infarction, heart failure, arrhythmias, cardiomyopathy, and vascular disease, while enabling evaluation of drug efficacy and toxicity. Although species differences require careful interpretation, rodent heart models provide accessible, mechanistic platforms for connecting molecular changes with whole-organ cardiac outcomes.

Isolated working heart models can be used to measure the effect of loading conditions, heart rate, and medications on myocardial performance and oxygen consumption. We describe methods for preparation of a rodent left heart working model that permits study of systolic and diastolic performance and oxygen consumption under various conditions.

Hemodynamic Characterization of Rodent Models of Pulmonary Arterial Hypertension

Pulmonary arterial hypertension (PAH) is a disease of pulmonary arterioles that leads to their obliteration and the development of right ventricular failure. Rodent models of PAH are critical in understanding the pathophysiology of PAH. Here we demonstrate hemodynamic characterization, with right heart catheterization and echocardiography, in the mouse and rat.

The Inverted Heart Model for Interstitial Transudate Collection from the Isolated Rat Heart

2017

This protocol describes a method to collect cardiac interstitial fluid from the isolated, perfused rat heart. To physically separate interstitial transudate from coronary venous effluent perfusate, the Langendorff perfused heart is inverted, and the transudate (interstitial fluid) formed on the cardiac surface is collected using a soft latex cap.
