Cell Pellet Preparation

Cell pellet preparation is a laboratory method used to collect cells from a liquid suspension into a concentrated mass for analysis or further experimentation. The process typically uses centrifugation, in which rapid spinning generates a force that moves cells toward the bottom of a tube, forming a pellet while the surrounding liquid remains above as the supernatant. Researchers can then remove the supernatant and resuspend, wash, lyse, or preserve the pellet, depending on the study. This basic technique supports cell counting, microscopy, molecular biology, protein analysis, culture maintenance, and sample preparation across biological research.

Liquid-grown Streptomyces cultures are characterized by mycelial pellets that are heterogeneous in size. We here describe a method to analyze and sort such pellets in a high-throughput manner. These pellets can be used for further analyses, which will provide leads to understand and control growth heterogeneity.

2016

In this study, a protocol was developed to produce good quality pellets using a flat die pellet mill at reduced specific energy consumption testing high-moisture corn stover and a starch based binder. The results indicated that adding a corn starch binder improved the pellet durability, reduced percent fines and decreased specific energy consumption.
